HOXB4 is a homeodomain-containing transcription factor with diverse roles in embryonic development and the regulation of adult stem cells. It promotes proliferation of hematopoietic stem cells and can both activate and repress apoptosis. Function: Sequence-specific transcription factor which is part of a developmental regulatory system that provides cells with specific positional identities on the anterior-posterior axis. Subcellular Location: Nucleus. Similarity: Belongs to the Antp homeobox family. Deformed subfamily. Contains 1 homeobox DNA-binding domain. SWISS: P17483 Gene ID: 3214 OXB4通過(guò)激活不同的信號(hào)通路增強(qiáng)HSC的自我更新,同時(shí)又不影響維持正常穩(wěn)態(tài)造血的調(diào)控機(jī)制。提高HOXB4的表達(dá)水平,能夠極大地增強(qiáng)HSC的自我更新功能,但基本不影響細(xì)胞的分化、系特異性及終末細(xì)胞的形態(tài)和功能。另外，HOXB4還可增強(qiáng)胚胎干細(xì)胞(embryonic stem cell,ESC)的造血潛能,促進(jìn)ESC向造血細(xì)胞分化。 |

Paraformaldehyde-fixed, paraffin embedded (Mouse placenta); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (HOXB4) Polyclonal Antibody, Unconjugated (bs-3650R) at 1:200 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.

Protocol
The cells were fixed with 4% PFA (10min at room temperature)and then permeabilized with 90% ice-cold methanol for 20 min at-20℃. The cells were then incubated in 5%BSA to block non-specific protein-protein interactions for 30 min at room temperature .Cells stained with Primary Antibody for 30 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.
